Job description

Human Technopole is a new interdisciplinary research institute, created and supported by the Italian government, whose aim is to develop innovative strategies to promote human health and aging through a multidisciplinary approach that integrates genomics, neuroscience, structural biology, computational and data science.

The Human Technopole Foundation is looking for a Senior Manager of gene editing pipelines to join the Automated Stem Cell and Organoid Facility (ASCOF). The aim of the Facility is to enable patient-specific iPSC-based disease modelling at scale by automatizing reprogramming, gene editing and organoids generation through the integration of state-of-the-art robotic and bioengineering platforms with a highly multidisciplinary approach. This includes translating, benchmarking and standardizing laboratory protocols into automated workflows, operating in close connection with the Light Imaging and Image Analysis, Genomics, Flow Cytometry and Electrophysiology facilities/support units to realize Human Technopole vision of high-definition biomedicine.

The successful candidate will report to the Head of Automation and focus on setting up the genome editing branch of the Facility.

S(he) will have the following main responsibilities:

  • Establish and manage the core functions of the gene editing unit, integrating general needs within Human Technopole and translating them into high quality operations and services;
  • Establish and standardize currently available gene editing technologies (plasmid DNA, mRNA, gRNA and Cas9 RNP) in pluripotent stem cells and cell lines;
  • Test and benchmark new technologies for the automation of the gene editing pipelines;
  • Oversee and assist with experiment validation and problem solving;
  • Design and run gene editing experiments in synergy with other members of the Facility;
  • Develop innovative strategies in genome engineering;
  • Keep detailed records of engineered cell line production and inventory;
  • Supporting the Head of the Facility in setting up the essential resources, participating in key aspects of general facility management and organization (e.g., managing inventories and stocks of samples, reagents, equipment and materials).

Job requirements

  • At least 5 years of relevant work experience is required:
  • Track record in the following areas is preferable:
    • Experience with the complete portfolio of CRISPR based genome editing techniques: from Knockout line generation to engineering of specific mutations and/or reporters, as well as gene expression manipulation through CRISPRi and CRISPRa, and CRISPR screening approaches;
    • Basic knowledge with prime-editing and base-editing methods;
    • Proven experience with mammalian cell culture techniques.
  • Familiarity with basic molecular biology and biochemistry methods (DNA and RNA manipulation, cloning, RT-PCR, immunoblotting and immunoprecipitation techniques) is preferable;
  • Experience and skills in performing repetitive and specialized laboratory experimental procedures and experience following experimental protocols is preferable;
  • Fluency in English – HT is an international research institute;
  • Ability to manage competing priorities in a fast-paced environment and to work independently;
  • Strong interpersonal skills demonstrated by the ability to lead and gain the assistance and cooperation of others in a team endeavor;
